Okay so you've never used Assos chamois cream before maybe because you don't use their shorts or in some belief that such products are for past times, but in doing so you have missed a real gem.
Way before the modern synthetic chamois was invented all shorts had a leather based outer to the pad which made contact with your skin. In the same way that leather shoes need to be treated to keep them supple and clean so you had to treat the leather chamois. The most popular products were various body moisturisers, as long as they had some lanolin content. Not only did this treat the chamois but it also reduced the friction against the skin, which resulted in a more comfortable ride.
Leather may no longer be used for chamois, but that doesn't mean there's no longer any use for chamois cream. You can use this product in two ways. First, treating the chamois - in doing so you are allowing the pad to soak up all that lubrication and store it for release throughout your ride. I have found this to an absolute godsend especially after five hours in the saddle nearing the end of a Sportive ride. The second way, and probably the most frequent way I use it, is to put the cream directly onto the skin. Due to the menthol in the cream it can leave the applied area feeling cold, not so bad in the heat of summer but less enjoyable in the colder months. This cold sensation soon wears off leaving the treated area protected for whatever the elements may throw at you.
Having used this product for over a year now both in Assos and other branded shorts I can't believe how simple yet effective it is. With ingredients such as Almond Oil (to soften the skin) Witch Hazel (cleansing, healing and soothing) every angle has been covered to improve a rider's comfort. I would recommend anyone to try this product; it could be the best money you spend all year.

I would just apply before the race.
I only use this on my road bike, but can say it is fairly greasy, so it does stick to your skin really well, but if it were me, I would apply it right before you hit the bike to get the maximum performance from it, unless your butt cheeks rub together too much when swimming :-)
Its seriously good stuff.. I used it twice now, and twice I have had zero sore bottom syndrome!
It says on the tub where not to put it.

Having just received a tub of this from Wiggle I suspect that they are shifting old stock as mine also lists Methylisothiazolinone as an ingredient (among others).
Google it then try and find someone who has new stock without this ingredient.
There are a lot of ingredients listed, with complex chemical names... I'm too lazy to type them all out for you.
Pop into a local bike shop, or buy it and make use of free returns if it's not suitable

Firstly, Vaseline, or another petroleum jelly provides excellent protection for nipple burn (having experienced this pain in the past). Either that, or elastoplast over the nipple can help by providing a barrier between your skin and the vest / tshirt.
Howeover, my primary recommendation, and something which I do myself as a runner, is to wear a lycra based compression top underneath my vest, or just wear that. This has totally eliminated the issue of nipple burn. I recommend Nike Pro products for this.

What sort of mileage do you get from a tub? Assuming you did a 90-100k cycle a week.
I lather quite a lot - probably 3 tubs a summery
We cycle 80-120 and it is ok. any more and you may be looking for some extra, but this may be all creams?
Never had to reapply even after rides of 10+ hours in 30 degree heat.
As I cycle to work and back, some times as much as 30 miles each away, I do apply it before each run.
MY GF however does take a lille pot to reapply after she has been to the loo - but men shouldn't need to reapply.
I would imagine if you let the cream get caked up within the pads that may cause some problems for the pads, and your bits!
What it does is prevent little infections from bacteria buildup and prevents any sore spots from chaffing or rubbing if you're riding for 5+ hours.
There's a slight menthol style cool tingle on your skin a few minutes after application. It's a remarkably pleasant sensation in case that sounds horrible.
