When installing a 10-speed cassette other than CS-4600, install the included 1.85 mm low spacer and the 1.0 mm spacer
included with the cassette at the positions indicated in the illustration.
For CS-4600, install only the 1.85 mm low spacer. (The 1.0 mm spacer is not necessary.)
Assuming the same holds true for other SRAM 10 speed cassettes then the answer provided by pkar of Melbourne is wrong.
To my Engineering mind: Use a thinner tyre. A lighter tyre, a more aerodynamic tyre. if you want a softer ride then use a slightly lower pressure. Finally puncture protection is more a component of the tyres makeup i.e. construction and not width.
So if you agree with this analysis, then of course go for the thinner tyre.
I have a Look 555 Ultegra with 2009 American Classic 420 34mm wheels. Would the Dura Ace 9000 C24 be significant upgrade? I am looking for good climbing and escending performance
Also a LBS owner advised me "Shimano don't place weight limits on their wheels. Many brands do, but most are less durable and more flimsy than Shimano's. C24's if you are over 115kgs I wouldn't personally use them. If you weight less than that I wouldn't think there would be any concerns".
They would be suitable for your requirements.

Rear = 829 g inc. rim tape
Total = 1425 g inc. tape, which makes the stated weight of 1395 very close!
Skewers = 61 g + 65 g
