In my experience personal comfort determines the quantity required. Through complacency I skimped on the quantity one day and soon learned to regret it while on the trail. Apply too much and things become mobile and a nuisance.
I did six hours last Saturday, from one application when the shorts went on.

Definitely before the ride.
You want to be spreading it on your "sit area" (inner thighs, pirineum and maybe between "glutes") so that you get good coverage and (if necessary) also applying some to the chamois inside the shorts itself.
Use as much as you need (I tend to scoop out a good dollop on my two first fingers).

I slapped a good amount on before, I haven't used it afterwards. when I dud get saddle sore I used sudocreme (nappy rash creme) which worked very well.
I used about twice the quantity of shaving foam you might use. this is probably too much but I'm not sure thats a problem, well it wasn't for me

Absolutely no problem. It tends to wear off the skin by the end of a very long ride (75 miles) but soap and water will remove it anyway.
I was my shorts on a 40 wash which removes it easily.

None at all in my experience - after a decent ride, its not visible on clothing and washing takes care of it, and as far as yourself is concerned, it comes off completely in the shower. In fact, you'll hardly notice it is there before the shower.

I don't think this product (or any other chamois cream) is gender specific. My wife uses this stuff on longer rides but prefers not to for anything under 50-60 miles though I would wear it to ride the length of our drive!

As far as I know, it should be fine. Although you should always be careful when putting creams near your nether regions, but that applies to personal hygeine as much as anything else (as in wash your hands before applications). Otherwise, just check against the ingredient list for allergies and perhaps do a test on an area that's not so sensitive. but otherwise get a removable chamois and cover it in cream and add this down your shorts at T1. I have done this for ironman's before. or do as I do now and just throw some bib shorts with a big cycling chamois (not triathlon chamois) over the top of my tri pants with thinner chamois.
but depends on distance of tri and if a westsuit swim. No wetsuit = cream will be useless, short tri then time in T1 is limitted. good luck

I wouldn't choose this particular product, go for a thicker one for a bit more staying power. I'd reccommend the new bodyglide chamois stick for max endurance. The udderly smooth didn't perform well even with a bit of rain whereas I've found that the bodyglide was still doing its job after 4 hours riding through a series of downpours!

The cream is waterproof and will last through a swim but it does depend on the length of the swim and the areas it is applied to. The cream is recommended for swimming so I think you should be fine during the riding stage of the event.

From my understanding this product originates from the dairy industry and was used to stop irritation of cows udders during milking. So from my understanding it has not only been tested on animals but they have benefitted from it for years before we got the opportunity.
Having used it myself I can confirm that no cows would have suffered irritation thanks to this product.
